Is there any possibility I can use the 2445j in place of a 2420 in a DIY 4333 build?

I would if I had aluminum, Be, or aquaplas dusted diaphragms in the 2445s. Stock, I'm not sure if I would prefer the change.
You obviously will need to change the horns, 2311 horns should drop in, but larger horns can work in DIY land. Do not use a pair of 2327 adapters.

You would have to change to a HL 93 2392 2" throat horn. Physical fit?? Probably. Might work out well. The horn in the stock 4333 is quite long.

I've used dozens of 2445s over the decades and made thousands of people happy with them. They aren't something I'd want to listen to in my living room. TADs in the day were much better sounding 2" drivers and B&C today makes much better sounding drivers. EV DH1s ( I guess thats heresy here ) are not bad either and will mount on any horn that fits a 2445. Putting an 8" mid cone between the 15 and the 1" HF driver would make more sense to me but then it wouldn't be a 4333.
I have the 2445's and am just trying to keep from shelling out the cash for the 2420's
