I just purchased a Marantz 2270 and a pair of JBL L100's. Someone on another board suggested that I run inline fuses between the two. Is this necessary/suggested?

If the gear doesn't have speaker protection and is know to go DC on occasion, it's a wise move.

Would I use a fuse holder like below on all four speaker cords along with 1.5 amp fast blow fuses, based on the L100's 50 watt rating??

That would work if you want to mount something. There's also options like this: http://www.napaonline.com/Search/Detail.aspx?R=BK_7822001_0282424653#
You only need it on the + side line.
Fuse size = square root of speaker max continuous power/speaker impedance
I=√50/8
=2.5A so I'd probably go 2A to be safe. 1.5A may be popping if you push it a lot.
Also remember the L100 ratings were conservative and standard different then so they'll take more "modern" rated power.
